Protein-ligand Complex Co-Crystallization & 3D Structure Determination

By SARomics Biostructures AB

We speed up our clients’ drug discovery projects through streamlined workflows and an extensive library of over 600 FastLane off-the-shelf drug-target proteins, which are ready for crystallization with new ligands. This allows for efficient 3D structure determination with a quick turnaround time. Additionally, our close proximity to the 4th-generation MAX IV beamlines further enhances our rapid processing capabilities. We also conduct ligand-binding assessments and interaction studies using 1D and 2D NMR spectroscopy. Protein-Ligand Complex Structures In SBDD X-ray crystallography is essential for detailed atomic-level analysis of protein-ligand complex structures in structure-based drug design (SBDD) and, more generally, in structural biology research. This requirement also applies to later stages of drug development, such as lead optimization and candidate drug development. SBDD aims to optimize ligand-protein interactions to achieve higher potency. This process usually runs in cycles (design-make-test-analyze). It requires many high-resolution (preferably better than 2.5 Å) ligand-protein complex structures (see our blog post on industrial use of synchrotron radiation in drug discovery). However, before obtaining the 3D structure of the protein-ligand complex, the target protein must be cloned, expressed, purified, characterized, and crystallized. This process can be time-consuming when starting from scratch, extending the early phase of drug discovery. Any acceleration of this process could help save time, cut costs, and enhance the project’s competitiveness. To accelerate our clients’ projects, we offer FastLane™ Premium and FastLane™ Standard off-the-shelf libraries that provide 600+ verified drug targets that can be directly crystallized with your chosen ligand (FastLane™ Premium) or expressed, purified, and crystallized according to verified protocols (FastLane™ Standard) within a few weeks. Further acceleration can also be achieved by choosing the right project strategy. In our technology section, you can find a discussion of the different strategies for SBDD. For details of crystallization and structure determination workflow, please visit our X-ray crystallography services page. For our fragment screening capabilities, please visit our drug discovery services page. Our laboratories and offices are located near MAX IV, a state-of-the-art fourth-generation synchrotron facility in Lund. We also offer priority access to the X-ray crystallography beamlines at MAX IV, BioMax, and MicroMAX, enabling rapid data collection and structure determination, helping ensure the success of our clients’ projects.
