GATTA-PAINT Nanoruler for higher temperature measuremnts DNA-PAINT measurements are typically performed on microscopy platforms at room temperature (21–25 °C). However, certain advanced systems (e.g., ONI Nanoimager, ONI Aplo Scope, among others) are equipped with temperature-controlled stages that operate at elevated temperatures (29–31 °C). These conditions significantly influence DNA-PAINT kinetics, accelerating the binding and unbinding rates of imager strands and thereby compromising the detection of reliable localization events. To address this challenge, we offer specialized GATTA-PAINT HEAT nanorulers, engineered for temperature-optimized performance. These standards enable robust and reproducible measurements under elevated stage temperatures, ensuring consistent DNA-PAINT imaging quality across a wider range of experimental conditions. product properties of GATTA-PAINT HEAT 80 nm and 40 nm Nanoruler - PAINT 80R HEAT: Color red; Available fluorophores ATTO 655; Recommended laser source 625 – 665 nm; Surface density ≈ 1/µm²; Distance 2x 80 nm; Level Beginner; Price 365,-€ - PAINT 80Y HEAT: Color yellow; Available fluorophores Cy3B; Recommended laser source 560 – 590 nm; Surface density ≈ 1/µm²; Distance 2x 80 nm; Level Beginner; Price 365,-€ - PAINT 40R HEAT: Color red; Available fluorophores ATTO 655; Recommended laser source 625 – 665 nm; Surface density ≈ 1/µm²; Distance 2x 40 nm; Level Beginner; Price 365,-€ - PAINT 40Y HEAT: Color yellow; Available fluorophores Cy3B; Recommended laser source 560 – 590 nm; Surface density ≈ 1/µm²; Distance 2x 40 nm; Level Beginner; Price 365,-€

The method STORM or dSTORM is one of the most common used super-resolution techniques in light microscopy. With the nanorulers from the GATTA-STORM series now the ideal calibration samples for this technology are available. To emulate your STORM or dSTORM measurements as facsimile as possible the nanorulers carry multiples of the common used dye Alexa647. We offer the mark-to-mark distances in the sizes 30 nm, 50 nm and 94 nm. On request we can also design special solutions for your specific requirements. All nanoruler samples will be delivered in solution (sufficient for at least ≥10 surfaces). Please be aware of that samples in solution demand your expertise and chemical facilities and that immobilizing and buffering requires additional chemicals, which are not part of the GATTAquant product. Recommended buffering for dSTORM is based on glucose oxidase, catalase and β-mercaptoethanol (see Rust, M. J.; Bates, M.; Zhuang, X. Sub-Diffraction-Limit Imaging by Stochastic Optical Reconstruction Microscopy (STORM). Nat Meth 2006, 3, 793–795). Further GATTAquant recommends the abbelight smart kit for measuring GATTA-STORM products. Nanorulers from the GATTA-PAINT HiRes series serve all advantages of the classical GATTA-PAINT nanorulers. Additionally, they include special DNA-PAINT based fiducial markers for a precise and reliable drift correction. These fiducial markers outclass common fiducial markers based on fluorescent beads by having a stable connection to the surface and emitting a signal which does neither bleach nor exceed the detector’s dynamic range. Nanorulers from the GATTA-RESI series serve all advantages of the classical GATTA-PAINT nanorulers and bring them to the next level of resolution. They are based on RESI (Resolution Enhancement by Sequential Imaging), a DNA-PAINT technique in which neighbouring docking sites carry orthogonal sequences and are imaged one after another. By combining all localizations of each docking site, the localization precision is improved far beyond conventional DNA-PAINT. Each GATTA-RESI nanoruler carries six pairs of docking sites, with the two sites of each pair placed 6 nm apart. The first site of every pair is imaged with the imager F1-LB, the second one with the imager F2-LB. Additionally, the nanorulers include DNA-PAINT based fiducial markers addressed by the imager F3-LB, which allow a precise drift correction and the alignment of both imaging rounds. For data analysis we recommend the open-source software Picasso. For GATTA-RESI we offer a mark-to-mark distance of 6 nm in the color yellow (Cy3B). The imagers F1-LB, F2-LB and F3-LB are included and supplied in solution. On request we can also design custom nanorulers with smaller distances for your specific requirements. product properties of GATTA-RESI Nanoruler: RESI Color: yellow Available fluorophores: Cy3B Recommended laser source: 560 – 590 nm Surface density: ≈ 1/µm² Distance: 6 nm Level: High-End Expert Price: 680,-€ High-End Expert: Samples which demand perfectly aligned microscopes and very experienced DNA-PAINT users which are able to optimize acquisition and data evaluation. fluorophore properties Fluorophore: Cy3B Our notation: Y Excitation / nm: 560 Emission / nm: 571

AFM images can be influenced by the form of the tip. These distortions can be corrected by using our GATTA-AFM 3D nanoruler as a calibration tool with a well-defined and known morphology for blind tip reconstruction. The design with sharply defined steps of different heights and a height profile well matched for use with a broad range of macromolecular complexes (up to 20 nm). It combines several characteristics that make it well-suited for blind tip reconstruction: - Its designed structure contains flat faces in the x-y direction and sharp edges in the z direction, creating a four-step staircase from 2 to 20 nm. - Consisting of DNA, it is fully biocompatible and can be facily deposited on surfaces alongside other biological macromolecules and imaged both dry and in liquid. - The above combination of features enables the nanoruler's application for straightforward AFM image correction for a wide range of 3D nanostructures. The samples are shipped in solution. The amount of material is sufficient for at least 10 AFM surfaces, densely packed with DNA-origami structures. Reference: Kolbeck, P. J., Dass, M., Martynenko, I.
