F-MRPS Viral Vector Quantification

By Spectradyne Particle Analysis

Accurate viral vector quantification: Concentration, size & nucleic acid content. Efficient cell transduction for gene therapy applications depends critically on high quality viral vector preps. But because virus is produced in complex biological systems, direct quantification poses a challenge. Live biological titer can take days to perform, and surface binding methods such as SP-IRIS or ELISA-based protein measurements do not provide accurate concentration. Mass photometry fails for impure samples, which can contain overwhelming concentration of non-viral particles. Fast & Easy: Virus Size & Concentration with MRPS. The figure above demonstrates the repeatability of MRPS for viral titer measurements. Human adenovirus 5 was measured in three technical replicates, showing a clear virus population in the sample with mean diameter approximately 85 nm and excellent repeatability. Each measurement required just a few minutes from start to finish, and no need for tissue culture or incubation. Virus purity and titer in a single measurement. In the example above, intact human adenovirus 5 (hAd5) are directly quantified within a complex mixture of off-target biological nanoparticles, in minutes, without tissue culture. hAd5 was mixed with a broad size distribution of extracellular vesicles, and the mixture was stained with a membrane-permeable intercalating dye to illuminate the nucleic acid contained inside the virus. With a single measurement of the stained sample, the concentration of the intact hAd5 is measured to be 5.4 x 10 9 particles/ml, representing 64% of all particles in the measured size range. Quantify a broad range of viruses with F-MRPS. Is lentivirus more your vector of choice? How about a vector from the Rhabdoviridae family? Here are a few different viruses effectively quantified with Spectradyne’s technology: - SARS-CoV-2 - Adenovirus - Lentivirus - Adeno-associated virus (AAV) aggregates - Bacteriophage (PR-772, T4, T3, L-VIR) - Influenza - Rabies - Maraba virus - Herpes Simplex Virus (HSV) - Human Immunodeficiency Virus (HIV) - Murine Leukemia Virus (MLV) - Mouse Hepatitis Virus (MHV) - Myxoma Virus Don’t see your favorite viral vector on this list?
